## Definition
**Iterative molecular staining** images multiple molecular targets by repeating cycles of labeling, acquisition, and signal removal or replacement in the same specimen. The registered cycles create a higher-dimensional molecular record than a single staining pass can carry.
## Applications in Use
[[wiki/PRISM (E11 Bio)|PRISM]] uses iterative staining to read combinatorial protein identities and synaptic markers from expanded neural tissue. Those channels supply identity and molecular context to the reconstruction model.
The preprint's contribution statement attributes iterative [[wiki/MAGNIFY|MAGNIFY]] work to [[wiki/Sung Yun Park|Sung Yun Park]] and [[wiki/Sven Truckenbrodt|Sven Truckenbrodt]]. This is a method contribution within the collaboration, not evidence that either person alone developed the full PRISM platform.
## Forward Capability
The governing variables are cycle count, registration error, tissue damage, signal carryover, marker specificity, and total acquisition time. Reliable cycles can turn an anatomical trace into a molecularly annotated connectome.
